Jentadueto

Jentadueto offers an advantage over monotherapy by combining two medications with different but complementary mechanisms of action. Linagliptin enhances the activity of incretins, hormones that reduce postprandial blood glucose levels. Metformin acts on the liver to reduce gluconeogenesis, thus lowering fasting blood glucose. Together, they help improve glycemic control without causing weight gain, a common side effect associated with some diabetes medications.

Description

Jentadueto is an oral medication used in the management of Type 2 diabetes. It combines two antihyperglycemic agents: linagliptin, a dipeptidyl peptidase-4 (DPP-4) inhibitor, and metformin hydrochloride, a biguanide. Linagliptin works by increasing hormones that stimulate insulin release in response to high blood glucose levels, while metformin decreases glucose production by the liver and its absorption in the gut. This dual mechanism helps control blood sugar levels in Type 2 diabetes patients. Jentadueto is available in tablet form for administration.

Additional information

Usage

Jentadueto is indicated for:

● The improvement of glycemic control in adults with Type 2 diabetes mellitus.
● Use in patients who are not adequately controlled on a regimen containing metformin or a DPP-4 inhibitor.
● As a second-line therapy when diet and exercise plus a single antidiabetic agent do not provide adequate glycemic control.

Patients are advised to:

● Adhere to their healthcare provider's instructions regarding dosing and administration of Jentadueto.
● Continue a proper diet and exercise regimen concurrent with medication use.
● Monitor their blood sugar levels regularly as advised by their healthcare provider.

Administration:

● Jentadueto is taken orally, with meals to reduce the gastrointestinal side effects of metformin.
● The tablets should be swallowed whole and not crushed, broken, or chewed.

Precautions:

● Jentadueto carries a boxed warning for lactic acidosis, a rare but serious complication caused by metformin accumulation.
● It should not be used in patients with renal impairment or metabolic acidosis, including diabetic ketoacidosis.
● Use should be temporarily discontinued in patients undergoing radiologic studies involving intravascular administration of iodinated contrast materials.

Drug Interactions:

● Alcohol intake should be limited as it can increase the risk of lactic acidosis when taking metformin.
● Certain medications (e.g., carbonic anhydrase inhibitors, corticosteroids, thyroid products, and others) may require more frequent blood glucose monitoring and may necessitate dose adjustments of Jentadueto.

Side Effects

● Nasopharyngitis (common cold symptoms).
● Diarrhea.
● Nausea and vomiting.

Serious side effects that require medical attention include:

● Lactic acidosis, especially in patients with kidney problems.
● Pancreatitis.
● Severe allergic reactions.

Storage

● Store at room temperature between 68°F and 77°F (20°C and 25°C).
● Keep the tablets in the original container with the lid tightly closed.
● Protect from moisture and keep out of the reach of children.

Special Precautions

Before taking Jentadueto, patients should inform their healthcare provider if they:

● Have any allergies to linagliptin, metformin, or other medications.
● Have a history of pancreatitis, kidney disease, liver disease, or heart failure.
● Are pregnant, planning to become pregnant, or are breastfeeding.

Overdose

An overdose of Jentadueto may lead to severe complications such as lactic acidosis or hypoglycemia. Symptoms of overdose may include weakness, lethargy, respiratory distress, or unconsciousness. In case of a suspected overdose, immediate emergency medical attention is necessary.
